Four New Types Developed for SEC and HILIC Chromatography |
TOKYO, Aug 31, 2016 - (JCN Newswire) - Showa Denko ("SDK"; TSE:4004) will start selling next month four new types of Shodex(TM) high-performance liquid chromatography (HPLC) columns, namely, "GPC HK" and "PROTEIN LW" packed columns for size exclusion chromatography (SEC*) as well as "HILICpak(TM) VC-50" and "HILICpak(TM) VN-50" packed columns for hydrophilic interaction chromatography (HILIC**).
"GPC HK" represents environment-friendly gel permeation chromatography (GPC***) packed columns that enable a very quick analysis and significantly reduce the volume of organic solvents used, mitigating the work load of molecular weight determination of polymers. Furthermore, "PROTEIN LW" and "HILICpak(TM) VC-50" enable high-sensitivity analysis of antibody medicine and substances in the brain, while "HILICpak(TM) VN-50" meets the need of high-sensitivity analysis in the area of bioenergy.
HPLC analytical columns are required to meet changing needs and their applications are increasingly diversified and sophisticated. SDK will continue to expand the lineup of high-performance products that will contribute toward industrial development in many areas, including pharmaceuticals and energy.
SDK will introduce the four new types of packed columns at JASIS 2016 exhibition (Japan Analytical & Scientific Instruments Show) to be held at Makuhari Messe, Japan, from September 7 through 9.
1. GPC HK Series for very quick analysis
While GPC (organic SEC) is the most commonly used method for molecular weight determination of polymers, SDK has been working to reduce the volume of organic solvent (used as eluent) and shorten the analysis time out of consideration for the environment.
The newly developed GPC HK Series has enabled us to shorten the analysis time to one-sixth that of conventional column (KF-804L) for the same range of molecular weight determination and reduce the amount of organic solvent to one-sixth. This is because the packing material, with smaller particle size, is uniformly controlled. Furthermore, it is not necessary to use different GPC columns for different types of solvents due to a high solvent exchangeability of GPC HK Series columns.
Due to the uniform particle size of the packing material, the pressure can be restricted to a low level even at high current speed. Thus, no analytical device for UHPLC is needed despite the quick analysis achieved by GPC HK Series.
2. PROTEIN LW Series for antibody medicine analysis
SDK offers PROTEIN Series gel filtration chromatography (GFC****) columns, which are aqueous SEC columns using silica-based packing material for protein/enzyme analysis. The newly developed PROTEIN LW Series is highly effective in separating antibodies, which serve as main ingredients of high-profile antibody medicine.
An antibody is a kind of protein that recognizes a specific foreign matter upon its invasion into the body, and connects with the antigen to discharge it to the outside of the body. Meanwhile, monoclonal antibodies (used in antibody medicine) tend to cohere. Dimers and aggregates, formed as a result of cohesion, change their activity and can cause side effects. Thus, it is important to analyze polymers in active pharmaceutical ingredients from a viewpoint of quality control of antibody medicine.
The PROTEIN LW Series has been developed through improvement of existing KW-800 Series columns - that have acquired good reputation for use in insulin analysis and other applications. Using our proprietary technology pertaining to surface modification of packing materials, we have enhanced separation efficiency of dimers and monomers, in particular.
3. HILICpak(TM) (two types)
SDK offers HILICpak(TM) Series columns, which enable high-sensitivity analysis of hydrophilic compounds. HILICpak(TM) VG-50, the first of the Series launched in 2014, improved the recovery percentage of reducing sugar, although it was an amino column. HILICpak(TM) VT-50, announced last year, enabled high-sensitivity analysis of anionic compounds, including saccharides (phosphorylated saccharides), organic acids, and various drugs. This time, we have developed HILICpak(TM) VC-50 that enables high-sensitivity analysis of cationic biological compounds and HILICpak(TM) VN-50 suitable for high-sensitivity analysis of oligosaccharides. We can now analyze a wider range of hydrophilic compounds with our HILICpak(TM) Series columns.
(1) HILICpak(TM) VC-50 for analysis of neurotransmitters In the newly developed HILICpak(TM) VC-50, carboxyl groups have been introduced into packing material. The columns enable high-sensitivity analysis of amine-based neurotransmitters and cationic compounds, including choline, acetylcholine, and amino acids. Different substances in the brain can be analyzed simultaneously using trace amounts of compounds. Thus, the columns are suitable for brain science research, involving neurological disorders.
(2) HILICpak(TM) VN-50 for analysis of oligosaccharides This is an HILIC column suitable for high-sensitivity analysis of oligosaccharides. Highly hydrophilic packing materials develop an excellent HILIC mode, realizing LC/MS analysis that was difficult with conventional SEC mode. Oligosaccharides are used in functional food and as raw material for bioethanol. As oligosaccharides are generated in the saccharification process of biofuel, it is important to analyze oligosaccharides in improving the efficiency of alcohol generation. It is now possible to fully separate various oligosaccharides in the saccharification process, using LC/MS analysis.

After separation of respective components of samples by liquid chromatography, separated components are detected by a mass spectrometer.
